The enzyme 'GalNAcEXO' is an α-N-acetylgalactosaminidase that efficiently hydrolyzes terminal GalNAc on glycoproteins. GalNAc, which is bound to serine or threonine and referred to as the Tn antigen, is rapidly and efficiently hydrolyzed by GalNAcEXO, and it also shows activity against α1-3 linked GalNAc. Introducing Genovis enzymes useful for the evaluation and analysis of antibody-drug conjugates. ◎FabRICATOR (IdeS) An enzyme that digests specific sites just below the hinge region of antibodies. It generates a uniform pool of F(ab')2 and Fc/2 with a 30-minute incubation. Typically, IgG is digested within 30 minutes, but there is no risk of excessive digestion even with longer incubation times. The enzyme activity is maximized at 37°C, but it can also be used at room temperature by extending the reaction time. ◎IgGZERO (EndoS) An enzyme that hydrolyzes the N-linked glycans in the Fc region of IgG. It is useful for reducing complexity through rapid deglycosylation of IgG and inactivating the effector functions of the Fc region. ◎GlycINATOR (EndoS2) An enzyme that hydrolyzes the glycans in the Fc region of IgG.
